Cadets Escherichia coli Gastrointestinal Infection Microbiological Contamination Milk Quality Most Probable Number (MPN)

Abstract

Background: Introduction: Milk is a nutritious food product that requires strict quality control to ensure safe consumption. Cadets at the Indonesian Defense University routinely receive milk as part of their nutritional enhancement program; however, its microbiological safety has not been comprehensively evaluated. Contamination by Escherichia coli may indicate fecal contamination and pose a risk of gastrointestinal infections.

Purpose: To analyze the presence of Escherichia coli in milk distributed to cadets for potential risk of gastrointestinal infections.

Method: This study used a descriptive analytical design. Milk samples were collected from male and female cadet dormitories over three weeks using a stratified time-point sampling method. Microbiological examination was performed using the most probable number (MPN) method, followed by confirmation through culture on eosin methylene blue agar (EMBA) and Gram staining. Data were analyzed descriptively and presented in tables.

Results: Escherichia coli contamination was detected in the first and second weeks, with the highest value being 7.2 MPN/ml. However, contamination decreased and was undetectable by the third week. Although contamination levels remained below the critical threshold, these results indicate inconsistent sanitation during milk storage and distribution.

Conclusion: Milk distributed to cadets is considered safe for consumption, but improvements in handling and sanitation practices are needed to reduce the potential risk of gastrointestinal infections. Routine monitoring is recommended to maintain microbiological safety.

Suggestion: Further research is recommended to conduct a more in-depth study of factors influencing microbiological contamination during milk distribution and storage, such as environmental sanitation conditions, storage temperature, and cleanliness of equipment used.
